Metformin has been the go-to medicine for managing Type 2 diabetes for more than 60 years. But it turns out it might be doing a lot more than just keeping blood sugar controlled. New research suggests the drug actually gets to work inside the brain by switching off a protein called Rap1.
Most of us have always thought Metformin worked by helping the liver produce less glucose and making the body more sensitive to insulin. However, as per a latest report in Science Alert, a team at Baylor College of Medicine has found something new: the drug seems to target a specific part of the brain that looks after our energy levels and sugar control.

The study showed that Metformin shuts down a protein in the brain known as Rap1. This protein is a bit like a manager for how the body handles energy. By turning it off, the drug helps the body regulate its metabolism in a way that scientists hadn’t actually realised until now.

The researchers also noticed that a particular group of brain cells, called SF1 neurons, sparks into life when Metformin arrives. These cells live in the brain’s main control centre for metabolism and seem to be a big part of how the drug manages our energy and glucose.

Why is this a big deal?

This is a big deal because it shows that Metformin isn’t just working on our organs like the liver or muscles, it’s actually influencing the brain’s own control systems. This could lead to brand new treatments that target these brain paths directly to help manage diabetes better.

Beyond just controlling sugar, Metformin has been linked to a few other health perks. Past studies have hinted it might help slow down brain ageing, protect our DNA, and even help people live longer. It’s even being looked at as a way to delay some of the downsides of getting older.
